Next, as has been observed, nature repeats itself by creating similar clonotypes that appear to have the same function 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, 17, 18, 19, 20, 21, 22, and these might be combined into groups. A clonotype can therefore be treated as the minimal functional group of antibodies. All antibodies within a clonotype-a group of antibodies that share a common ancestral recombined cell that arose in a single donor-usually perform the same function. Nevertheless, some inferences can be made. Innovative methods such as mitochondrial lineage tracing could perhaps be used to validate computed clonotypes 26. However, in the absence of a sufficiently large dataset with multiple antigen specificities using cells from multiple humans or donors, it is currently impossible to assess the validity of any functional grouping scheme. In the future, antibody properties might be understood at scale from sequence information alone, perhaps via structural modelling, which could lead to antibody grouping 25. Larger numbers of antibodies can be assayed for simple binding to a particular antigen. In practice, small numbers of antibodies are assayed in vitro–for example, for functional activities such as neutralizing capability. Ideally, antibodies in such groups would share both an epitope and complementary paratopes dictated by their protein sequences. Similar content being viewed by othersĪ central challenge of immunology is the grouping of antibodies by function. For most functional antibodies, the heavy chain determines the light chain. Thus, although naive antibodies seem to recur by chance, the recurrence of functional antibodies reveals surprising constraint and determinism in the processes of V(D)J recombination and immune selection. We also observe this phenomenon when similar heavy chains recur within a donor. This property of functional antibodies is a phenomenon that we call light chain coherence. We show that for naive antibodies (those not yet adapted to antigens), the probability that they use the same light chain V gene is around 10%, whereas for memory (functional) antibodies, it is around 80%, even if only one cell per clonotype is used. Here we consider pairs of B cells that share the same heavy chain V gene and CDRH3 amino acid sequence and were isolated from different donors, also known as public clonotypes 23, 24. Each chain contains three complementarity-determining regions (CDR1–CDR3), which contribute to antigen specificity. The charter in question, pictured below, is among the oldest in the college’s possession and dates from the mid-12th century (reference Brackley B.186). To illustrate what this might entail, and to show why I personally find charters (and large collections thereof) so engaging, I thought I’d spend my first contribution to this blog looking in detail at just one of the items still housed in the Muniment Room. But, as Magdalen’s own collection makes clear, modern historians and others can still call on thousands upon thousands of charters to inform their research. Of course, survival rates come nowhere close to this number. ( Michael Clanchy, for example, calculated that the peasant classes alone produced millions of charters in the century up to 1300.) They were, therefore, everyday objects produced en-masse with everyday implications. This would have also been true for charters in the early medieval period, when such things were typically issued (and retained) by those at the very top of medieval society.īy the 13th century, however, charters were being produced by members of almost every social rank, such that modern estimates for how many were written involve some fairly eye-watering numbers. After all, Magdalen can lay claim to some fairly spectacular medieval items in its collections, including a beautifully illuminated copy of Bede’s Ecclesiastical History and a manuscript once owned by Thomas Becket, both of which I was lucky enough to see during a recent student teaching session hosted by Dr Hannah Boston in the Old Library just before my arrival. Some of you may be thinking that this is hardly surprising. Also frustrating are the 3 or so “Unread Direct Messages” you can see, when you tap “Show All” it takes you to the “Direct Messages” sidebar that sorts in its own way, with unreads strewn about. Seems like the “Dark Mode” folks won the design argument as the new app no longer supports the desktop or web app color themes which now makes the iPad app incredibly difficult to know at a glance which team you’re viewing. Now, the iPad app has been turned to be quite bland. No longer easy to see all unread messages in one place - which was a feature I regularly requested for the desktop app. In some ways, it is almost as if Slack doesn’t understand its users. The recent iPad App update misses the mark quite considerably. One Step Forward, Two Steps Back for iPad We'd be happy to investigate the issues you are having with editing and sending messages. Luckily it’s miles better than Discord, which although more powerful, is not easy to use or grasp. Right now it doesn’t ‘feel’ right - I know that doesn’t really help but perhaps they’d benefit from a larger user-acceptance phase for UX. I also would like to see a more user friendly search query system (AND/OR) where I can add criteria. I would like to see a more clear distinction between ‘search everywhere’ and ‘search within the current channel or private conversation’. Lastly, I think they need to rework ‘search’. I also find the app to respond slow on iPadOS, as if it’s not native but some kind of ‘HTML shell’ that doesn’t really bring the best of the native CPU/GPU capabilities. I know Slack used to be a game company, but this is not the type of game I’m interested in playing :) I had to move it out of the way, and quickly hit ‘submit’ before the bar jumped back, a matter of milliseconds. But this button is hidden behind a tiny floating language selection & dictation bar. When pressing ENTER on the iPad keyboard, the message is not sent you have to press a button.
